Lysosomes are essential cellular organelles known to be the main site for\ud the breakdown and recycling of endocytosed macromolecules within cells.\ud However, our knowledge of lysosomal function has changed considerably\ud over the last decade with the lysosome now being a recognized Ca2+\ud signaling organelle with additional roles in plasma membrane repair,\ud clearance of defective organelles and mediating cell death as well as\ud established roles in clearing infection. Our ability to study these unique\ud organelles has however been stymied by a dearth of good techniques for\ud the purification of functional lysosomes not contaminated by other\ud organelles.
