The cryopreservation of the ovarian preantral follicles could help the conservation of several domestic and wild animal species. The objective of this investigation was to verify the effect of the ethylene glycol in different concentrations in nuclear organization de ovarian preantral follicles. Ovaries had been gotten in slaughter house. A toxicity test was conducted with strips of ovarian cortex using ethylene glycol (10, 20 or 40%). Tissues analysis were run using classic techniques of histology. The analysis of the morphologic organization of the nuclear chromatin of primordial follicles showed significant difference (P < 0.05) between the control group (81,1%) and 10% (60,5%), 20% (19,3%) and 40% (9,5%) groups. In primary folículos, significant differences between the control group and 20% (18,7%) and 40% (9,5%) groups (P < 0.05), but not between group 10% (65,5%). These results indicate that the cryoprotectant ethylene-glycol takes the morphologic alterations in the nuclear chromatin of primordial and primary follicles, enclosed in bovine ovarian tissue.
A criopreservação de folículos ovarianos pré-antrais pode ajudar na conservação de muitas espécies domésticas e selvagens. O objetivo do presente estudo foi verificar o efeito do etileno-glicol, em diferentes concentrações, na organização morfológica da cromatina nuclear de folículos pré-antrais inclusos em tecido ovariano bovino. Ovários foram obtidos em abatedouro. O teste de toxicidade foi realizado com fragmentos ovarianos expostos ao etileno-glicol em concentrações de 10, 20 ou 40%, diluído em solução de NaCl a 0,9%. O tecido foi analisado por técnica histológica clássica. A análise da organização morfológica da cromatina nuclear de folículos primordiais, mostraram diferença significativa (P<0,05) entre o grupo controle versus grupos 10%, 20% e 40%. Em folículos primários, foram observadas diferenças significativas entre o grupo controle e grupos 20% e 40% (P<0,05), mas não entre o grupo 10%. Estes resultados indicam que o crioprotetor etileno-glicol leva a alterações morfológicas na cromatina nuclear de folículos primordiais e primários, inclusos em tecido ovariano bovino.
